EXOSOMES ARE PERFECT THERAPEUTIC VECTORS

TARGET SPECIFIC CELLS
TO TRIGGER A PRECISE CURATIVE ACTION

Ciloa’s technology targets therapeutic cell exosomes on specific cells to trigger a precise curative action.

Exosomes are natural nano-carriers, perfect for traveling in the human body for delivering new messages. Ciloa’s technology allows to naturally load exosomes with 2 types of proteins :

  • the first one, at the surface, to target the diseased organ;
  • the second one, as a cargo inside the exosome, to deliver a signal (for modification, multiplication or death).

The best analogy is a guided missile that recognizes its target and modifies it.

Exosomes can fuse back with cell membranes, introducing new proteins and RNA in new cells. An increasing body of evidence shows that exosomes are natural intercellular shuttle vehicles of materials and of information that can reprogram targeted cells. They are able to pass through any body barriers, including the Blood Brain Barrier.

Ciloa’s genetically engineered exosomes will provide novel innovative vectors for drug delivery.

PROVEN EXOSOME PLATFORM FOR SPECIFIC TARGETING & DELIVERY

A) Fluorescent regular exosomes (control exosomes) or

B) fluorescent customized exosomes (exosomes with a ligand) harboring a ligand protein specific of a chosen neuron receptor were incubated with neuronal cells in culture.

Customized exosomes harboring the ligand are captured with very high efficiency by neuronal cells, to be compared with the very low level of capture of regular exosomes devoid of the chosen ligand. These results reveal that our customized exosomes deliver specifically their cargo in the targeted cells.

CONTROL EXOSOMES
≤ 5 % capture
EXOSOMES WITH A LIGAND
≥ 95 % capture

TECHNOLOGICAL CONSTRAINTS

Currently, gene therapy generally uses vectors based on virus or synthetic nano-materials that present some constraints such as :

  • Immune rejection
  • Possible toxicity
  • High clearance and poor kinetic
  • Unability to pass through the Blood Brain Barrier
  • Low target specificity

OVERCOME TECHNOLOGICAL LOCKS

Benefits to use Ciloa’s recombinant exosomes :

  • Ability to transport drugs to targeted organs
  • Passage of all barriers (including the blood brain barrier)
  • Avoid the use of viruses (potentially dangerous & immunogenic)
  • Avoid the use of artificial nano-materials
  • Stability, biocompatibility, no toxicity
  • Specific interaction with cells; phenotype modifications

Exosomal membrane is robust enough to ensure the protection of RNA/gene of interest against any enzymatic digestion.

More, after release they can interact with new cells and fuse back to their membranes, delivering new proteins to these cells. This phenomenon can be used to target specific cells with particular proteins. Thus, Ciloa solution can be used, for example, to target cancer cells with exosomes harboring toxic molecules or molecules recruiting the immune system.
